Outline of Annual Research Achievements |
In the past one year, I have performed deep transcriptome sequencing on Megabalanus rosa cyprids. The transcriptome assembly of M.rosa cyprids contains 144690 sequences. In silico peptide mining was performed on the transcriptome assembly, yielding 11 putative neuropeptide and 15 putative peptide hormone precursors. We also produced antibody for in-depth analysis of predicted peptide hormones.

Reason
We have acquired nice assembly of M.rosa cyprids, which has aided the accurate in silico peptide mining step greatly.

Strategy for Future Research Activity |
I am now synthesizing short peptides belongs to three well studied families, allatostatin, tachykinin and sulfakinin. We will test their functions in isolated M.rosa cyprids brain once we receive these short peptides. The in vitro experiments will be a pilot testing. If we get clear-cut results, i.e. distinguishable reaction upon application of short peptides, we will synthesize more peptide hormone/short peptides to build a comprehensive view on the role of neuropeptides on larval settlement. We will also perform immunostain and western blot to study the protein expressions of peptide hormones.

Causes of Carryover |
The first year, I focused on transcriptome sequencing and bioinformatic analysis. The assigned budget for last year is more than enough to cover the expanse on transcriptome sequencing step. Based on the transcriptome results, we bought antibodies for in-depth analysis. I will need to use the rest of the money to cover the next important step, the peptide synthesis step.
