Outline of Annual Research Achievements

ヒトCCD患者由来iPS細胞(CCD-iPS細胞)のほか、遺伝子編集操作によりRUNX2変異を正常化したiPS細胞(Revertant-iPS細胞)の樹立に成功し、これを報告した(Stem Cell Research & Therapy (2018) 9:12)。さらに同様の方法でノックアウトiPS細胞(KO-iPS細胞)を樹立し、これらの細胞を骨芽細胞へと分化誘導する過程において、RUNX2欠損細胞が核溝や分葉核といった異常な核形態を示すことを発見した。また、核構造維持に関連する遺伝子群の発現を調べたところ、RUNX2欠損細胞では核形態維持に必須のLamin A/C遺伝子発現が著しく低下していた。以上のことから、CCD病態におけるRUNX2発現・機能減弱と核形態維持に関わる遺伝子発現に関連がある可能性が考えられた。
