本科研費初年度末より、The Ohio State Universityに研究代表者が1年間滞在し、ヒト乳児由来糞便細菌叢移植gnotobiotic pigsを用いた栄養充足（sufficient diet, SD）群と、栄養不足（deficient diet, DD）群におけるロタウイルス（RV）ワクチンの効果検証実験を行った。最終年度は得られた実験の統計解析を進めた。結果、DD群はワクチン投与後の強毒株の攻撃に対して下痢防御率が低く、胃腸炎スコアは攻撃後4日目において、SD群より有意に高かった。ウイルス排泄量は全般的にDD群で高く、経過を通じた幾何平均として有意差が認められた。血清中のグルコース値は、強毒株攻撃前後ともに、SD群で有意に高かった。また、LPSは強毒株攻撃当日の測定において、DD群で有意に高かった。血清中の抗RV IgA抗体は両群で強毒株攻撃後に上昇したが、攻撃後の値はSD群のほうが有意に高かった。糞便移植2日目のIL-22がDD群で有意に低く検出された。単核球フローサイトメトリー解析では、ワクチン投与試験において、安楽殺時の脾臓由来HRV特異的IFN-γ産生性CD4+T細胞の存在率はDD群で、有意に少なかった。これらの結果から、食餌中のタンパク欠乏により、IL-22の低下で示される腸上皮細胞のバリア機能の破綻および、LPS高値の示すBacteremiaがもたらされた可能性が示唆された。また、抗RV IgA抗体の産生不良や、RV抗原で刺激した際のT細胞のIFN-γ産生能が低下していることから、ワクチン接種に対する免疫応答が阻害され、結果的にRVワクチンの効果減弱を引き起こしていることが示唆された。ヒト乳児由来糞便細菌叢移植gnotobiotic pigの系は、腸内細菌叢、栄養と免疫との相互作用解明に有用な動物モデルであり、RV感染症対策には、ワクチンの活用に加えて、衛生や栄養も考慮した包括的な取り組みが必要なことが示唆された。
