Research Abstract

本研究は、発がん防御という視点に焦点を絞り,転写因子機能を総合的に理解することを目的としている。本年度は、血球前駆細胞の増殖・分化・アポトーシスの制御を介して赤血球および巨核球の形成過程において重要な転写因子GATA1に着目し、マウス個体を用いた解析を行った。近年、N末端領域を欠失したGATA1変異が、ダウン症候群患児に発症する一過性骨髄増殖症(TMD)や、ひきつづく巨核芽球性白血病(AMKL)の芽球にほぼ必発していることから、自血病発症にかかわるGATAI変異の関与が注目されている。私たちは、同様なGATA1変異をもつマウスでは、TMD患児と同様に、胎生期から新生児期に未熟な巨核芽球が著明に蓄積しているが、この表現型は出生後すみやかに消失することを見いだした。また、ヒト症例と異なり、成獣マウスでは白血病を発症しないことを見いだした。このことは、GATA1単独変異によりTMDが発症しうるが、AMKLの発症には、21番染色体トリソミーに起因する他の遺伝子異常が引き金となることを示唆している。私たちは以前、GATA1ノックダウンマウスの解析を通して、GATA1の発現量の低下により、赤血球前駆細胞が蓄積し、その中の一部のマウスが前赤芽球性白血病を発症すること報告した。これらの結果は、GATA1の量的異常(発現量の低下)と質的異常(構造異常)が、それぞれ系列特異的な未熟な前駆細胞の蓄積を招来し、白血病発症の素地を作ること、さらに、それぞれの白血病発症には、GATA1変異にさらなる未知の遺伝子変異が関与している可能性を示唆している。
