がんの発生と進展におけるDNAメチル化およびヒストン修飾異常の役割

Research Abstract

DNAメチル化やヒストン修飾異常の解析が進みつつあるが、腫瘍の種類によっては、エピジェネティックに不活化を受ける標的遺伝子の情報は十分ではない。乳癌および多発性骨髄腫においてエピジェネティックな異常により不活化されている遺伝子の網羅的解析を試みた。その結果、これらの腫瘍でDNAメチル化により不活化されている新規遺伝子を同定出来た。乳癌において、NTN4遺伝子のメチル化は高頻度かつ癌特性が高く、診断のマーカーとして有用な可能性が示唆された。また、多発性骨髄腫で異常メチル化されているRASD1遺伝子は、デキサメサゾン感受性に関与し、薬剤感受性を予測する分子マーカーとして有用である可能性が示唆された。これまで、マイクロアレイを用いたゲノム網羅的エピゲノム解析を行ってきたが、non-coding RNAに対するプローブがないため、既存のアレイではゲノムのカバー率が不十分であった。抗ヒストンH3K4Me3抗体でクロマチン免疫沈降したDNAを次世代シークエンサーで解析することにより、約25,000の新しい遺伝子転写開始点を同定した。その情報を用いて、DNAメチル化により不活化される新規microRNAや機能性RNAを同定した。これまで、大腸癌のプロファイルにより、一部の癌ではゲノムワイドなDNAメチル化異常を示すことを明らかにしてきた(CpG island methylator phenotype, CIMP)。これらの腫瘍の前癌病変は、BRAFやK-ras変異を有し、細胞老化に関する遺伝子のエピジェネティックな不活化を有すること、大腸拡大内視鏡で特徴的な表面形態を示すことを明らかにした。
