がん組織における内因性血管新生抑制のシグナリング機構

Research Abstract

Vasohibin-1のがん組織における発現の解析を進め、乳癌において癌細胞の増殖や生存期間から判断して悪性のものほど腫瘍血管での発現率が高いことが判明した。Vasohibin-1のELISAによるアッセイ系を構築し、がん患者の血中Vasohibin-1濃度を測定したところ、肺癌患者において術後血中Vasohibin-1濃度が有意に低下することが観察されたが、肺癌、子宮癌、卵巣癌の患者にっいて血中Vasohibin-1濃度を健常者との間で比較では、有意の変化は観察されなかった。
Vasohibin-1の機能を知るためVasohibin-1 KOマウスにルイス肺癌細胞を移植したところ、移植腫瘍は腫瘍血管新生に富んでおり、内因性Vasohibin-1は腫瘍血管新生確かに制御していることが確認された。腫瘍を移植した野生型マウスにヒトVasohibin-1遺伝子搭載非増殖型アデノウイルスを尾静注すると、外因性Vasohibin-1は腫瘍血管を成熟化させた。また、その結果として血流が改善し、抗癌剤との併用効果が観察された。
Vasohibin-1ホモログであるVasohibin-2は、血管新生局所に浸潤する骨髄由来単核球に発現し、Vasohibin-1と拮抗して血管新生を促進することを観察した。そこで癌組織にっいて検討したところ、癌細胞もVasohibin-2を発現していることを観察した。そこでVasohibin-2発現陰性の腫瘍細胞にVasohibin-2を導入してマウスに移植すると、腫瘍血管新生と腫瘍発育が増強し、逆にVasohibin-2発現陽性の腫瘍細胞のVasohibin-2をノックダウンすると腫瘍血管新生と腫瘍発育が抑制されることが判明した。
