ストレス応答シグナルによるがん細胞の増殖・死の制御

Research Abstract

本研究計画は、細胞の分化と死のシグナル伝達機構の機能解析に焦点を絞ったものであり、その成果は癌化メカニズムの理解に大きく貢献するものと考えられる。特に細胞の分化ならびに死に関わる細胞内シグナル伝達機構を明らかにすることを目標として設定した。昨年度に引き続き、本年度も、ASKファミリー分子群の機能解析を中心に研究を遂行し、ストレス応答MAPキナーゼ経路の制御機構の破綻と発がんとの関連性の解明を試みた。二段階皮膚発癌モデルを用い,ASK1ならびにASK2ノックアウト(KO)マウスにおける皮膚腫瘍の形成について検討した。マウスの背部にイニシエーターとしてDMBAを塗布後,プロモーターとしてTPAを継続塗布して経過を比較したところ,ASK2KOマウスにおいては,野生型マウスに比べて形成された腫瘍数が明らかに多く,また発生時期も比較的早いことが明らかとなった一方で、ASK1KOマウスにおいては、TPAによる炎症の誘導が低減しており、野生型マウスと同程度にしか腫瘍が形成されなかった。DMBA処理後の皮膚におけるアポトーシスをTUNEL法にて検出したところ,ASK1ならびにASK2KOマウスにおいてDMBA処理によって生じるTUNEL陽性細胞が減少している傾向が認められた。以上の結果より、ASK1ならびにASK2はDMBAによるアポトーシスの誘導に必要であり、ASK2が新たながん抑制遺伝子としての機能を有することが判明した。一方で、ASK1は、アポトーシスの誘導と炎症誘導に重要な機能を持つことが明らかになった。
