Research Abstract

血管抑制タンパク・トロンボスポンジン-1(TSP-1)が、従来の報告と異なりCD36非依存性に内皮細胞の細胞周期を停止させる作用のあることを見出した。すなわち、CD36のほとんど発現しないヒト臍帯静脈内皮細胞において、TSP-1により非リン酸化Rbとp21タンパクの上昇が観察され、apoptosis誘導ではなくG1停止が引き起こされた。VEGFR1からのシグナルは、血管内皮のみならず単球・マクロファージの遊走に重要であることを我々は明らかにしてきた。これについてさらに解析するため、VEGFR1高発現のヒト血管内皮細胞由来細胞株AG1-G1-Flt1を樹立した。この細胞株、および、複数のVEGFR1変異cDNAを強発現する細胞株もちいて解析し、PLC・-PKC-MAPキナーゼ経路とは独立の細胞遊走シグナルがカルボキシル末端領域から発信されることを明らかにした。また、VEGFR1シグナルはマクロファージ系を介してリンパ管新生を引き起こすことを、VEGF-AおよびVEGF-EトランスジェニックマウスとVEGFR1-TK-/-マウスをもちいて示した。VEGFR2からのシグナルを単独で検討できるVEGF-Eについて、創傷治癒など組織再構築に有用であることを見出した。さらに、ヒトへの応用が可能なヒト型化VEGF-Eの大量調製系をCHO細胞で構築した。発現の程度については、さらに改良の余地があり、現在、検討を進めている。
