Research Abstract

我々は、癌の本質が無制限・無秩序な細胞増殖である点に着目し、細胞増殖動態を正確に診断し得る細胞周期プロファイル解析法を開発した。本法では、CDKs1,2,4,6の特異活性、および、Cyclines(A,B,D,E)とCDK inhibitors(p16, p21,p27)のタンパク発現量を同時に測定することが可能である。今年度の研究目標は、この細胞周期プロファイル解析法を用いた乳癌の細胞増殖動態の解析とそれに基づく乳癌の予後および薬剤感受性診断法の開発である。我々は、まず細胞周期プロファイル解析法を用いて284例の原発乳癌の細胞周期を解析した。その結果、CDK2/CDK1特異活性比が高い乳癌は増殖が速く再発リスクが有意に高いことを見出した。更に、CDK2/CDK1特異活性比は、従来の病理学的予後因子およびエストロゲン受容体やHER2過剰発現とは独立した予後因子であることを明らかにした。また、パクリタキセル感受性乳癌細胞株ではパクリタキセル接触後24時間でCDK1/2比が増加するのに対して、耐性株ではこの比が増加しないことに着目し、パクリタキセル感受性診断法(乳癌組織より採取した乳癌細胞をパクリタキセル存在下に24時間培養しCDK1/2比を測定する)の開発を試みた。25例の臨床サンプルを用いて検討した結果、本法は、パクリタキセルの臨床効果を高い精度(正診率(92%))で予測し得た。以上の結果は、細胞周期プロファイル解析が乳癌の予後およびパクリタキセル感受性の診断に有用であることを強く示唆している。
