TGF-βシグナルを応用した制癌法の開発

Research Abstract

1. ヌードマウスに移植したヒトスキルス胃がん組織のDNA microarray解析を行い、TGF-βシグナルに応答して血管新生を抑制する遺伝子を同定した。すでにThrombospondin-1が血管新生抑制因子として働き、TGF-βシグナルを遮断すると脳rombospondin-1の発現が低下し、血管新生が亢進して腫瘍の増殖が見られることを明らかにしていたが,平成20年度はさらにTIMP-2がTGF-βシグナルの遮断により低下すること、この結果、腫瘍血管新生の亢進に関わっていることを明らかにした。またスキルス胃がん細胞からがん幹細胞(Cancer stem cell)をFACSを用いて単離することを試み,得られた胃がん幹細胞がTGF-βによって分化する可能性を示唆する結果を得た。さらにBMPシグナルを遮断した場合でも増殖が更新することを確認し、平成21年8月までにその分子機構の解析をDNA microarrayなどを用いて完了した。
2. TGF-βによって誘導され上皮-間葉細胞分化転換に関わる転写因子(SnailやSlugなど)と癌との関わりについて検討を行った。膵臓がん細胞Panc-1ではTGF-βによってEMTが誘導されるが、EMTの誘導には活性化されたがん遺伝子Rasのシグナルが重要であることが明らかとなった。TGF-βによってSnailが誘導されるが、Rasシグナルの存在下ではSnailの誘導が顕著に見られた。一方で,他のTGF-βの標的遺伝子の誘導はRasシグナルの有無でも大きな影響を受けず、Smad7はむしろRasシグナルの存在下では抑制されることを確認した。このことからTGF-βによるEMTの誘導にはRasシグナルとの協調作用が重要で、SmadシグナルとRasシグナルの間にがん治療の新たな標的分子が存在する可能性が示唆された。
