Research Abstract

ヒト脳腫瘍の中でも最も悪性度の高い膠芽腫の検体を用いて、TGF-βが脳腫瘍幹細胞の維持に寄与していることを突き止めた。さらにTGF-βの作用を種々の阻害剤で遮断することで、悪性脳腫瘍の増殖が抑制され、分化が誘導されることを発見した。さらに、TGF-βが脳腫瘍幹細胞の維持のさいに、新規のTGF-β下流遺伝子Sox4の発現を通じて転写因子Sox2の発現を誘導していることを見出した。この研究成果は脳腫瘍のがん幹細胞に対してTGF-β阻害剤やSox4、Sox2の機能抑制が有効であることを示すものであり、今後、臨床応用に向けて研究を発展させて行く。
TGF-βは種々の細胞に作用して、増殖抑制作用やアポトーシス誘導などにより細胞の増殖を抑えることが知られている。我々はTGF-βの細胞増殖に関連した第3の作用としてTGF-Bが肝臓がん細胞などに作用してオートファジーを起こすことを発見した。TGF-βによる増殖抑制やアポトーシスの少なくとも一部は、オートファジーによって誘導されていることを明らかにした。
スキルス胃がんでTGF-βシグナルを遮断するとin vivoでの腫瘍増殖が亢進した。またがん遺伝子c-Skiを発現することでTGF-β受容体の異常と同様の変化が起こることを明らかにした。さらに既に報告していた血管新生抑制因子Thrombospondin-1の腫瘍細胞からの産生の低下に加えてTIMP2などのマトリクス分解制御タンパク質が血管新生の亢進に関わっていることを発見した。
スキルス胃がん幹細胞をSide population分画およびALDH1発現の有無によって単離することに成功した。スキルス胃がん幹細胞はヌードマウスに移植する強い造腫瘍性を持ち、また自己複製することを確認した。脳腫瘍幹細胞と逆に、スキルス胃がん幹細胞はTGF-βにより造腫瘍性を喪失することを見出した。
