白血病細胞のシグナルと分子標的治療

Research Abstract

チロシンキナーゼの恒常的活性化は、各種造血器腫瘍の発症や病態に深く関わっている。申請者らは、c-kitレセプターチロシンキナーゼ(RTK)の恒常的活性化変異が腫瘍性マスト細胞や消化管ストローマ細胞腫瘍にも存在することを見出し、RTKによる腫瘍化機構とその制御法について検討している。本年度は、慢性好酸球性白血病の原因遺伝子FIP1L1/PDGFRαを中心に検討を行った。FIP1L1/PDGFRα遺伝子をレンチウイルスベクターを用いて未分化な形質を有するKit^<high>Sca-1^+ Lineage^-(KSL)造血幹細胞に導入すると、サイトカイン非依存性増殖分化能や自己複製能が誘導された。一方、FIP1L1/PDGFRα遺伝子を各種造血前駆細胞に導入した場合、サイトカイン依存性増殖は認められたが一過性であり、自己複製能はほとんど誘導されなかった。興味深いことに、FIP1L1/PDGFRα遺伝子をKSL細胞や骨髄系、巨赤芽球系、リンパ球系など各種造血前駆細胞へ導入すると好酸球への分化能が誘導されたが、TEL-PDGFRβやFIP1L1/PDGFRβ遺伝子を造血幹細胞や造血前駆細胞へ導入しても好酸球系への分化は誘導されなかった。FIP1L1/PDGFRαは、TEL-PDGFRβに比してMEK1/2やP38MAPKをより強く活靴し、C/EBPα、GATA1やGATA2の類も増強させた。さらに、FIP1L1/PDGFRαならびに下流に存在するRasはPU.1の転写活性可能を低下させることが明らかとなった。以上のことから、FIP1L1/PDGFRαが最も未分化な造血幹細胞に生じ、RAS/MEKやP38MAPKの活性化ならびに系統特異的転写因子の発現や活性を変化させることにより好酸球性白血病の表現系が生ずると考えられた。これらの知見は、今後の白血病幹細胞の特性解析に有用である。
