化学療法と抗体療法における細胞死の耐性機序の解析

Research Abstract

抗体療法や分子標的薬剤の細胞死における機序または耐性機序はひとつではなく、T315Iの点突然変異が有名であるが克服する薬剤を探求している。薬剤については耐性克服薬剤として、Src kinase inhibitor, proteasome inhibitor、HDACI、NF-kB阻害剤がある。これらの薬剤による克服が可能かどうかを検討した。In vitroではT315Iを克服できる可能性のある所見が得られており今後広く症例から細胞を得て、検索していく。今後抗体療法の耐性機序についてもさらに具体的な分子の解析が必要である。これらについては共焦点レーザー顕微鏡などを用いて測定系を作成しており、今年度は特に新たに見いだしたB細胞性リンパ腫におけるCD20抗原遺伝子の点突然変異をみるために、N末端に特異的な抗体を作成し、これまでに汎用されているL26抗体との比較、耐性例での検討を免疫染色、細胞内CD20蛋白の分布など解析した。CD22抗体医薬やシグナル伝達阻害についても研究した。またADCC測定系を確立したので、固形がんにおける種々の抗体医薬の活性を測定した。EGFR系の抗体医薬では、erbituxでEGFの完全阻害とADCCが測定された。KRASの野生、変異型についても検討中である。またSTAT3阻害剤、TKIの新たな耐性機序についても研究した。また骨髄腫についてはbortezomib耐性を作り、機序の研究を行った。
