Research Abstract

グルタミン酸受容体δ2(GluRδ2)は小脳プルキンエ細胞特異的に発現し、平行線維-プルキンエ細胞の後シナプス部位に局在している分子である。GluRδ2はシナプス可塑性(LTD)、シナプス形成と神経回路網形成及び整備に重要な役割を果たしている。しかしながら、GluRδ2がどのようにシナプス可塑性、シナプス形成、神経回路網形成・整備を調整しているのかは依然として不明である。GluRδ2のC末端(T site)にはPDZドメインを有するPSD-93、PTPMEG、Delphilin、nPIST、S-SCAMが結合する。GluRδ2のT siteを欠失したGluRδ2ΔTマウスは運動失調を示さず正常に発育した。GluRδ2ΔT蛋白は野生型GluRδ2蛋白と同様に平行線維シナプスの後シナプス肥厚部に存在し、GluRδ2ΔTマウスにおけるシナプスの密度、形態ともに野生型と比べ有為な差は認められなかった。しかしながら、GluRδ2ΔTマウスにおいては小脳LTD誘導の異常と運動学習の障害が観察された。さらに、GluRδ2ΔTマウスでは登上線維支配領域の遠位方向への拡大と遠位樹上突起において登上線維支配の異所転位が認められた。一方、発達に伴う近位樹上突起での過剰な登上線維の除去過程は正常におこなわれていた。このように、GluRδ2のT siteは小脳LTD誘導と登上線維支配領域の調整に必要不可欠である一方、平行線維シナプス形成と発達に伴う近位樹上突起での過剰な登上線維の除去過程には必要ではないことが明らかとなった。これらの結果は、平行線繊シナプスに局在すGluRδ2がT siteを介したシグナル伝達により登上線維支配領域の調整しており、これらの調整機構がプルキンエ細胞における平行線繊と登上線繊シナプス形成の異種入力線維間競合に重要な役割を果たしている事を示唆するものである。
