Research Abstract

本研究では, 新規ナノバイオ環境システム技術を確立するために, ナノマニピュレーションに基づいた局所環境計測・制御用バイオナノツールの創製し, これまで不可能であった局所的な環境情報の計測・制御技術, さらには低侵襲な細胞計測技術を確立することを目的としている.
昨年度に引き続き, 環境制御型電子顕微鏡における高精度・高作業性を有した多自由度多プローブナノマニピュレーションシステムを用いた各種バイオナノツールの創製・単一細胞計測実験環境を整備・応用している. 本年度は, バイオナノツールとして, ナノプローブを用いて, 単一のイースト菌の局所的な硬さ計測を行うために, ナノプローブの座屈変形に基づいた微小力計測手法を提案した, 実際に, 単一のイースト菌に対して局所硬さ計測実験を行い, 成長段階や観察条件に基づいた新たな知見を得た. また, マイクロ流路内に任意形状のマイクロ構造物を紫外線硬化樹脂により作製する手法を提案し, 本手法により作製したマイクロ構造物をレーザーマニピュレーションによりその場操作することにより, より複雑な回転体などのマイクロ機械構造物としてアセンブリを実現し, また駆動実験を行うことで, 特性を評価した. また, チップ内でエレクトロ・フォーメーション法により, リポソームを作製し, マイクロ・ナノピペットにより電気浸透現象を利用することで, 単一の任意のリポソーム内に生体試料を局所的に注入する技術を提案した. これによりこれまで困難であった分子量が大きく粘性を有するような生体試料を細胞と同様の閉鎖空間内で反応させるための新たな実験環境を提案した.
