体節の周期的分節パターン確立機構の解析

Research Abstract

脊椎動物の分節性(繰り返しパターン)の形成及び、分節境界の形成に関わる分子機構の解明を目指している。今年度は、Notchシグナルの修飾を介して分節時計や分節境界形成に重要な機能をもつことが知られている糖鎖修飾酵素であるL-fngの体節形成過程における必要性を検討した。L-fngの発現パターンは複雑で、未分節中胚葉の後方では、振動パターンを示し、これがNotchシグナルの負の因子として機能してNotchシグナルの振動をうみだす。一方、未分節中胚葉の前方では安定的な発現を示し、ここでの活性が体節の分節境界の形成に重要であると考えられていた。今回この2つの発現を分離できるトランスジェニックマウスを作成し、それらをL-fng欠損マウスと交配し、内在性のL-fngがない状態で解析し、どちらの発現がどのような機能をレスキューできるかを検討した。その結果、Hes7-プロモーターでドライブしたL-fng(すなわち、後方の振動をつくりだすL-fng)のみで正常な体節の分節及び体節の前後極性の確立に必要・十分であることが明らかになった。また前方のL-fngはNotchの切断には必要であることが分かったが、その結果分節境界に形成されるNotchの切断は分節境界の形成には必要ではないことが明らかになった。また我々はコンピューターシュミレーションを用いて、Notchシグナルの振動が分節境界の形成や体節の前後極性の確立に必須な分子であるMesp2の転写制御に必須であり、そのためにL-fngが機能していると結論づけた。
