| Research Abstract |
phot分子は,光屈性,葉緑体光定位運動,気孔開口光制御などを担う青色光受容体で,N-末端側LOVと呼ばれる発色団ドメインを二つ,C-末端側にセリン/スレオニンキナーゼドメインを持ち,光より制御されるタンパク質キナーゼとして機能すると考えられる。これまでにphotのLOV2ドメイン光反応機構を解明する一方,二つLOVドメインが,キナーゼの光制御おいて異なる役割を果たすこと示唆する結果を得ている。昨年度の研究によりシロイヌナズナphotでは,LOV2がキナーゼ活性部位光可逆的に結合して,活性制御の光分子スイッチとして機能すること,photlとphot2ではキナーゼ性制御のモードが異なっていることを示唆する結果を得た。さらにdocking simulationおよびアミ酸変異導入によりにより,phot2のキナーゼドメインとLOV2との結合サイトの詳細な解析を行った。方LOV1の機能として,キナーゼ活性の光制御の光感受性の低下作用,およびphot分子の二量体化サトの機能を持つのではないかという結果を得ていたが,昨年度,世界で始めてLOV1の二量体結晶構を明らかにし,LOV1がタイマーサイトとして働く事を証明した。この結果を得て,二量体構造のアステリックな効果による光感受性の制御の可能性を考察した。
