磁気的手法によるバイオイメージングと脳機能ダイナミックスの研究

Research Abstract

本研究は,脳や神経の局所的磁気刺激による神経活動の制御,及び新しい手法による神経活動の電流分布イメージングや脳波計測により,高時間・空間分解能を有する新しい脳機能ダイナミックスイメージング法を構築し,脳機能の動的機構の解明に迫る脳機能研究法の確立を目的としている.本年度は以下に述べるような成果を得た.経頭蓋磁気刺激時の脳内電気現象を知るために脳波との同時計測が有効であるが,磁気刺激時のアーチファクトが問題となっている.ここでは,アーチファクト除去のため独立成分分析を用いた信号処理を行うことにより,刺激後10msから誘発脳波の観測が出来,脳内電流分布の推定が可能となった.また,視覚認知情報処理を調べるため,視覚探索課題に磁気刺激を適用し,脳内情報処理め時間特性を調べた結果,視覚刺激提示後150msにおいて,右後頭頂葉が視覚探索処理に関わっていることがわかった.MRIを用いたバイオイメージングにおいては,脳における細胞膜機能に着目して,水分子に対する細胞膜の透過率をMRIの信号から推定する新しい手法を提案した.水分子の拡散と磁気共鳴現象を組み合わせた数値解析手法を開発し,MRIの信号取得に合わせてパルス傾斜磁場を加えたときの信号強度を,様々な細胞膜透過率に対して計算した.実験では,ラットの大脳皮質から選択的にMRIの信号を取得するのに合わせて,傾斜磁場を加えた.数値解析結果と実験結果とを比較することにより,未知だったラットの大脳皮質の細胞膜透過率が,74μm/secと求まった.この手法は,ヒトにも適用可能であり,脳活動に深く関係する細胞膜機能の基礎研究や,脳梗塞などの疾患の早期診断などへの応用が期待されている.
