Research Abstract

神経の電気活動によって生じる微弱な磁場を磁気共鳴イメージング(MRI)によって検出できれば,高い空間分解能および時間分解能で,脳機能ダイナミックスの計測が可能になる.本研究では,神経活動による磁場がMRIの信号に与える変化を,理論と実験の両面から評価した.神経活動を電流ダイポールによりモデル化して,その近傍に位置するボクセルの信号強度を解析したところ,ダイポールモーメント12nA・mに対して,0.6%の信号低下が得られた.また,ラットの左坐骨神経を電気刺激しながら,主磁場4.7TのMRI装置を用いて,刺激後30ms間隔で画像を取得した.右体性感覚野において,刺激後0-30msにおいて一時的な信号低下がみられ,これは神経活動に伴う微弱な磁場によるものと考えられた.また,磁気ナノ粒子を加えて,アミロイド沈着を促進させて,感度を上げて,アミロイドの沈着の様子をMRIで検出できるようにするための基礎研究という位地付けで,磁気ナノ粒子のコロイド溶液が数μg/mlの低い濃度でも可視光領域で蛍光発光を示すことを観測して,蛍光分光でナノ粒子水分子の動的挙動のサイズを測定し,βアミロイドへのナノ粒子の結合による機能化の判定や蛍光分子での測定限界内での凝固サイズの決定を行った.さらに、磁場印加群と対照群で,βシートアミロイド沈着の形成の比較を行い,磁気ナノ粒子と結合したアミロイドシートに交流磁場を印加したアミロイド溶液が赤外分光スペクトルのβシート領域でピークが小さくなることを見出した.このことは,本実験で,交流磁場印加により,凝固の割合が減少したことを意味している.さらに,経頭蓋磁気刺激(TMS)時の事象関連電位の計測に成功し,左縁上回に与えるTMSが事象関連電位P300に影響を及ぼすことがわかった.
