Research Abstract

DNAに種々の光機能性クロモフォアおよびナノ粒子を修飾した光機能化DNAを反応場として電荷分離・電荷移動系を構築すると共に、DNAのビルディングブロックとしての性質を巧みに利用し、光機能性をデバイス表面上に集積化し光電変換デバイスとしての応用へと展開することを目的としている。本年度は、修飾DNAにおける高効率光電荷分離系の構築を行うとともに、電荷分離収率とDNA損傷との関連性について検討を行った。光電荷分離収率の向上を目的として、光電子移動によって生じるイオン対の再結合が逆転領域の深い領域で起こると考えられるジフェニルアセチレン誘導体を修飾したDNAを用い、時間分解過渡吸収法によって電荷分離収率およびその機構に関する研究を行った。その結果、光照射によってマイクロ秒以上の寿命を持つ電荷分離状態を数十%の収率で生成し、光活性分子の酸化還元電位を制御することで高効率に電荷分離状態が生成することを明らかにした。さらにドナーとアクセプター間を架橋しているアデニン塩基に置換基を導入することで酸化電位を変え、レドックス勾配を与えることによって電荷分離収率が上昇することも明らかとなった。これまでに確立した光電荷分離系を光デバイスへと応用するために電極表面にDNAを固定化し、光電流測定による評価を行う計画である。また、酸化損傷機構の解明とPDTへの応用を目的とし、光電荷分離過程とDNA損傷の関係について検討した。光酸化損傷の効率は、光増感剤と損傷のターゲットであるグアニン塩基間の塩基対の数に依存することを明らかにし、グアニンの相補塩基であるシトシンに置換基を導入することで損傷効率を変化させることに成功した。損傷ターゲットとなる配列を詳しく調べた結果、アデニンが連続する配列を持つDNAでは電荷分離が促進され損傷効率が著しく増加することが明らかとなった。
