神経可塑性モデルとしての神経因性疼痛の発症・認識機構の体系的研究

Research Abstract

我々は、神経損傷1週間後でも、グルタミン酸NMDA受容体NR2Bサブユニットの1472番目のTyr残基(Y1472)のリン酸化、神経型一酸化窒素合成酵素(nNOS)の活性化による機能的変化により維持されていること、一酸化窒素(NO)が順行性・逆行性メディエーターが脊髄における神経可塑性に重要な役割をすることを明らかにしてきた。
今年度、慢性疼痛に伴う機能的変化について以下の興味ある知見が得られた。
1) Thy-1-YFPマウスを用いた神経再生
神経損傷に伴う神経変性・再生過程をin vivoで可視化するために、神経選択的に蛍光タンパクYFPが発現するThy-1-YFPマウスで神経再生モデルを確立し、軸索-Schwann細胞の相互作用を解明した。
2) 慢性疼痛の発症・維持機構の解明
(1) NOの下流のカスケードとしてcGMP/PKG系とS-ニトロシル化がそれぞれ神経伝達物質の遊離に関与し、脊髄後角での中枢性感作に関与することを明らかにした。
(2) nNOSのトランスロケーションを可視化するために、PDZドメインを含むN末端部分(1-299残基)とYFPの融合タンパクnNOSNT-YFP nNOSを用いてATPがnNOSの活性化に関与することを明らかにした。
(3) NR2Bのコンディショナルノックインマウスに神経因性疼痛モデルを作製後、脊髄後角を摘出しPSD画分の群(各140匹)間比較をプロテオミクスで解析し、機能分子を同定した。
(4) プロスタグランジンF2αとATPの機能的カップリング
3) PET用リガンドとしてNOS阻害薬の有機合成と評価、アクロメリン酸誘導体を合成した。
