Research Abstract |
In this study we analyzed the structural organization of two cortical regions, main olfactory bulb(MOB)and hippocampus in the C57BL/6J strain mice. In the hippocampus we performed quantitative analyses of chemically defined GABAergic neurons by means of a modern stereological method, optical disector using the con focal laser scanning light microscopy, and revealed their regional and laminar composition. Furthermore we performed the quantitative analysis of microglia and revealed their spatial distribution in the hippocampus. In the MOB we performed the immunohistochemical analyses at the light and electron microscopic levels and the tracer experiments, and revealed the followings. 1) We confirmed the compartmental organization of the glomerulus and the presence of type 1 and 2 periglomerular cells. ls and the vast majority of type 2 periglomerular cells(all calbindin- and 65% of calretinin-positive periglomerular cells)were GAD/GABA-positive. 2)The nitric oxide synthase(NOS)-containing neurons were heterogeneous, including some subpopulations of periglomerular cells, granule cells, interneurons in the external plexiform layer(EPL), superficial and deep short-axon cells and stellate cells. Importantly some NOS-positive large short-axon cells were projection neurons. In addition, some middle and external tufted cells were also NOS-positive, some of which constituted the intrabulbar association system and the projection system to the olfactory tubercle and the areas covering the dorsal endopiriform nucleus, claustrum and insular cortex. 3)By means of the retrograde tracer experiments some particular types, that is, the larger group but not the smaller group, of tyrosine hydroxylase-positive GABAergic juxtaglomerular neurons were revealed to be the main source of the long intraglomerular connections in the mouse MOB.
