| Research Abstract |
(1)炭素負イオンパターン注入による高分子基材上での間葉系幹細胞の自発的パターン配列
ガラス基板上のポリチレン薄膜や医療用シリコーンラバーシートに炭素負イオンをスリット列のマスクを介して注入処理を行い、ラット骨髄由来の間葉系幹細胞を播種して培養した結果、炭素負イオン注入のパターンに沿って、自発的な配列接着がいずれの高分子材料においても得られた。ポリスチレンでは最適注入処理条件を探査した結果、注入エネルギーが5〜20keVの範囲では3×10^14ions/cm^2の注入量が、また、10keVでは1×10^15ions/cm^2の場合が良好な自発配列が得られた。本条件での24時間水中浸漬後の接触角は約70度であり、親水化による選択接着と言える。しかし、接触角が60度程度となる注入量が更に多い場合は、間葉系幹細胞の接着は得られるがパターンからはみ出しており、パターニング性が悪い結果となった。他方、シリコーンラバーでは同様な注入エネルギー範囲では注入量が1〜3×10^15ions/cm^2とポリスチレンよりも多い場合が良好な自発的パターン配列接着を示した。これはシロキサン結合が炭素-炭素結合よりも強い結合エネルギーを有しているため、炭素負イオン注入処理では強い処理が必要となるためである。
(2)パターン配列した間葉幹細胞の神経細胞への分化誘導
間葉系幹細胞が自発的パターン並列接着を示した後に、βメルカプトエタノールを刺激剤とした2培地交換法による分化誘導を行った結果、パターン配列を維持したまま神経細胞に分化させることに成功した。神経細胞への分化確認は、神経特異エノラーゼの蛍光観察により行った。これにより、間葉系幹細胞を用いて、人為的な神経回路網形成を実現した。なお、容量性電極を組み込んだ基材上での間葉系幹細胞や分化した神経細胞への電気刺激は、電極の溶出などの問題で実施出来なかった。
(2)負イオン注入パターン化処理による脳神経細胞の人為的神経回路網の形成
脳神経細胞(ラット胎児大脳皮質神経細胞)のパターン配列では、ポリスチレン上にポリDリジンをコートし、パターン負イオン注入パターン処理を行った。その結果、ポリDリジンへの炭素負イオン負イオンではそのデグラデーションにより未注入領域に神経細胞を選択接着させることに成功した。
