| Research Abstract |
本年度は、以下の4点についての検討を重点的に行った。
1.IFNARの発現調節機構の解明
IFNAR遺伝子の解明について、methylationとその発現強度の関係の検討を目的として、methylation-spacific-PCRにより解析した。臨床検体での同じ検討により、methylationが機序の一つであるという感触を得た。
2.IFNARを介した5-FUの抗腫瘍効果増強作用の解明
5FUのDNA損傷作用へのIFN-alphaの作用について、臨床検体を用いたTS阻害率について解析した。この結果より、5FUの代謝からみた臨床症例における効果予測の可能性について、その他の因子についての絞り込みを行った。
3.death ligand(TRAILやFasLなど)を介した抗腫瘍効果の増強機構の解明
Fas-FasLを介したIFN-alphaによる宿主の免疫賦活作用と5-FUとの相互作用、特にapoptosis増強作用については、肝癌細胞株により、関与するものとそうでないものがあることが判明した。
4.治療抵抗性進行肝細胞癌症例における上記の基礎的検討項目とIFN併用化学療法の臨床効果との関係
cDNA microarrayを用いた網羅的遺伝子解析により、in-vivoモデルでの腫瘍検体をもちいて本療法の効果予測に関する遺伝子群についての解析を施行した。いくつかの可能性がある遺伝子群を臨床検体での解析結果と比較した。
以上の結果により、機序解明から効果増強、網羅的遺伝子解析から効果予測が可能である可能性が強くなった。今後の検討により、基礎的検討結果を臨床応用できると考えている。
