| Research Abstract |
【Bone invasion model mouse】 We conducted three experimental studies by using the bone invasion model mouse. 1) Tumors, five oral cancer cell lines(S AS, Ca9-22, HSC-2, HSC-3, HSC-4) were administered on periosteum with inflammation caused by LPS. 2) Tumors were administered on periosteum in the Osteopontin knock-out mouse. 3) Tumors were administered on periosteum that was irradiated. When the tumors were administered on periosteum, the bone invasion was not shown in each tumor. Therefore, B16 malignant melanoma cell that was the liquid cancer cell, was administered on the periosteum. The bone invasion was shown with the irradiation.
【Gene expression profiling】 The possibility that the function as a barrier to the tumor invasion fails by the irradiation was shown. Therefore, the periosteum may analyze the gene expression profile of the irradiation site. The extraction of mRNA was tried from the periosteum. Though, enough mRNA to supply it to microarray cannot be collected.
【Measures concerning bioethics and for safety】 After this research concerned is approved by institutional review board in June 2005, the cancer cells have been continuously gathered from the oral cancer patient who obtained informed consent in ethical review board in this organization. Patient's individual information is being managed as an anonymous data base by the network independent computer.
