Research Abstract

It has been demonstrated that obesity associates with increased breast cancer risk as well as a poor prognosis. In order to disclose the mechanism of adipocytokines for breast cancer initiation and progression, we investigated the expression of adiponectin receptor 1 (AdipoRl) and receptor 2 (AdipoR2) in the human breast epithelial cells and breast cancer cells. AdipoRl and AdipoR2 expression was recognized in both normal breast epithelial cells and breast cancer cells by immunohistochemistry. These results might indicate that adiponectin exerts its effects directly on normal breast and breast cancer cells.
Next, we examined the mRNA expression level of leptin receptors (long isoform; Lep-R(L) and short isoform ; Lep-R(S)) in breast cancer tissue. Both Lep-R(L) and Lep-R(S) high tumors were significantly associated with a poor prognosis. Furthermore, the significant association between both Lep-R(L) and Lep-R(S) mRNA high levels and a poor prognosis was recognized only in the subset of patients with high serum leptin level.
These results suggest a possibility that adipocytokines including adiponectin and leptin might modulate the growth of normal breast epithelial cells and breast cancer cells directly.
