Research Abstract

During this study period, we performed Hedgehog (Hh) signaling system analysis in 45 thymus tissues and reported analytical results about 26 specimens (Anticancer Research 25:00 3697-3702,2005). Summary of results obtained during the study period is as follows.
1. We analyzed for the first time Hh signaling system in the adult thymus tissue. As a result, Shh was frequently expressed in the adult normal thymus, but Glil, which is a marker of the activation of Hh signaling pathway, was not expressed.
2. Interestingly, Shh expression showed an inverse relationship to histological classification of WHO in the thymoma. Namely, Shh expression rate was 100% in type A,91.6% in type AB,62.5% in type Bl,25% in type B2,and 0% in type C. Glil expression was 100% in type A,36.4% in type AB,75% in type B1,25% in type B2,and 100% in type C. Thus, Glil expression was significantly higher in the thymoma than in the normal thymus (P<0.0001), suggesting Hh pathway activation in the thymoma.
3. Because Shh expression was hardly found in one thymic carcinoma, in which a marked Glil expression was found, ligand-independent activation was suggested in this case.
4. Twelve of the 26 cases were myasthenia gravis cases, but were not able to find significant relations between myasthenia gravis and Hh signal activation.
5. The establishment of the thymoma cell line did not succeed.
