Research Abstract

癌抑制遺伝子の欠失や癌遺伝子の増幅をともなう染色体変化は癌細胞の特徴である.また近年注目されている機能性RNAも癌細胞で発現の異常を示し,その背景に該当領域のゲノムのコピー数異常が存在することも示唆されている.遺伝情報の量的変動すなわち染色体あるいはその特定領域のコピー数変化を解析する技術としては,比較ゲノムハイブリダイゼーション法(CGH),代表差異分析法(RDA)および古典的細胞遺伝学的手法がある.CGH法など細胞分裂中期の染色体を利用する方法では解像度の限界が20Mb程度もあり,より小さな領域に限定されたコピー数変化の解析は不可能である.最近はCGH法からマイクロアレイへの移行によって解像度が増しているのが現状である.
本研究はヒトゲノムDNA量を高解像度かつ高精度に調べるシステムとしてデジタルゲノムスキャニング法(Digital Genome Scanning,以下DGS)を開発し,疾患ゲノム研究に実用化することをめざしている.DGS法はゲノムを制限酵素処理して得られる短いDNA断片配列をライブラリ化し,塩基配列解析と断片数の集計により,ゲノム上で異常な増幅や欠失を示す領域を同定する手法である.これまでの実験によって,in silico解析では(1)制限酵素処理で得られる全DNAバーチャル断片情報の収集とデータベース化,(2)コピー数異常同定のコンピュータシミュレーションによるDGSの実効性の確率論的検証を行ってきた.さらに,in vitro解析では胃癌細胞株ゲノムにおいてmyc(1Mb,10倍程度)およびk-ras(0.5Mb,6倍程度)癌遺伝子が存在するゲノム領域の増幅の同定に成功した.
