Outline of Annual Research Achievements |
In the final year of the JSPS fellowship (2018-2019), we established new tools and methodologies to address technical difficulties encountered during our research. Most significantly, we succeeded in optimizing buffer conditions for the deposition of ligated λ-DNA (~100 µm in length) on APTES-coated glass slides using the nanopipette with a counter electrode. Imaging of the patterned lines was done in a fluorescence microscope.
We successfully implemented new strategies to overcome significant resistant to flow of water and liquid metals as channel cross section decreases in diameter, from ~500 um to ~100 nm: 1. A new pipette holder was constructed, enabling simultaneous application of high pressure (up to ~200 Atm), heating (up to ~500 C) and high voltage (up to 10 kV) to pipette. Using the new holder, we were able to extrude GaIn alloy through the nanopipette channel. 2. Protocol for coating of the inside of glass capillaries with gold using electroless plating was established, with successful reproducible results. Gold coated nanopipettes will be used for various applications.
