Outline of Annual Research Achievements |
Project 1 - Dissecting physiological role of lncRNAs with SINEB2 elements: We have finalized the list of SINB2 containing lncRNAs (research plan; project2; a) that we want to dissect in details by analyzing different database analysis. We looked for their expression in different tissues and established cell lines and their involvement in different patho-/physiological functions. Moreover, we started with and got some initial data regarding research plan; project2; a) & b). Project 2 - SINEUP Sequence motifs and RNA binding proteins (RBPs): We finalized the protocol for ‘modified eCLIP’ in house. We also prepared the initial libraries for sequencing.

2: Research has progressed on the whole more than it was originally planned.
Reason
We added a new Aim-2 in order to address our original research question to dissect the mechanism of SINEUPs. So far, the research activity to address the new aim moving forward smoothly. I strongly believe, we can finish the projects on time.

Strategy for Future Research Activity |
Project 1: Dissecting physiological role of lncRNAs with SINEB2 elements 1) Defining relationship between IncRNA/SINEB2 and sense protein coding genes by knockdown and overexpression experiments, 2) Optimizing protocols (RNA FISH, ChRIP-Seq/MS etc) to evaluate the mechanism of action of IncRNA/SINEB2. Project 2: SINEUP Sequence motifs and RNA binding proteins (RBPs) 1) Finalize modified eCLIP protocol in house, 2) Generate biological replicate eCLIP library for validation, 3) Sequencing and data analysis, 4) Write manuscript and present our finding at international conferences.
