Research Abstract

光合成酸素発生系の反応機構、獲得過程、葉緑体への継承過程の解析のために研究を進め、以下の結果を得た。
(1)クロロフィルdを主要色素とするシアノバクテリアAcaryochloris marinaから、高い活性を保持する光化学系II標品を得る方法を確立した。さらにAcaryochloris淡路株について生理学実験に適用可能な培養条件の検討を進めると共に、光質条件(白色光と遠赤色光)を変えて培養した細胞についての時間分解蛍光スペクトルの解析を行い、光化学系2反応中心の電子供与体がクロロフィルaであることを明らかにした。
(2)酸素発生反応における中間状態遷移のpH依存性を、FTIR法により解析し、プロトン放出を起こす遷移を特定した。
(3)始原的シアノバクテリアGloeobacter violaceusの酸素発生系表在性タンパク質には、特異な欠失、挿入があるが、その影響は光化学系II中のCP-43、CP-47の変異により相補され、活性が維持されることを明らかにし、酸素発生系の多様性を初めて示した。
(4)第一電子供与体クロロフィルの高い酸化電位の要因を量子化学計算によって調べ、それが蛋白質内の極めて低い誘電率に起因することを示した。
(5)全ゲノム配列を用いた分子系統解析の方法を開発し、光合成生物に応用した。その結果、進化の過程で、まず緑色光合成細菌(光化学系I型)が誕生し、次に紅色イオウ光合成細菌(光化学系II型)が現れ、最後に酸素発生型光合成細菌であるシアノバクテリアが誕生したことを明らかにした。
(6)原始紅藻シアニディオシゾンの核ゲノムにコードされているシアノバクテリア由来の遺伝子のプロモーター領域の塩基配列を解析し、それらのうちのいくつかは、核ゲノムに散在しているくり返し配列に含まれるプロモーター様配列から転写されていることを明らかにした。これは、核ゲノムの散在性繰り返し配列が葉緑体から移行した遺伝子のプロモーター配列の起源になっていることを示唆する結果である。
