Outline of Annual Research Achievements

本年度は、主に分子スケールと細胞スケール、メゾスケール、マクロスケールの研究課題を推進した。主要な研究実績は以下の通りである。
1.分子モーターで駆動される微小管の数理モデル化に成功し、2本の繊毛が協調運動することで効率的に水流を生み出していることを明らかにした。(Omori, et al., J. Biomech. Sci. Eng. 2018) 2.繊毛虫のテトラヒメナが、固体壁面に留まることができることを実験的に示し、そのメカニズムを数値シミュレーションで明らかにした。この成果は、科学で最高峰のPNAS誌に掲載された。(Ohmura, et al. Proc. Natl. Acad. Sci. USA, 2018) 3.繊毛虫のゾウリムシも固体壁面に留まることができることを実験的に示し、細胞形状の影響を数値シミュレーションで解明した。(Nishigami, et al., Commu. Integr. Biol., 2018) 4.大腸菌が壁面近傍でどのように振舞うのか、数値シミュレーションで検討した。その結果、腸壁のようなひだが存在すると、壁面に長時間滞在できないことが明らかになった。(Yang, et al., Physical Review E, 2019) 5.マウス初期胚において、ノード繊毛が作り出す流れがどのような生物学的機能を生み出すのか、マルチスケールの数値シミュレーションを行った。その結果、センサー繊毛が水流を感知していることが示唆された。(Omori, et al., Roy. Soc. Open Sci. 2018) 6.遊泳微生物を模擬した、マイクロロボットの推進メカニズムを考案した。流体振動を制御することで、任意の方向に運動させられることを示した。(Morita, et al., Physical Review E, 2018a, b)

Reason

2018年度は、特筆すべき研究成果として、科学で最高峰のPNAS誌に論文を掲載することができた(Ohmura, et al. Proc. Natl. Acad. Sci. USA, 2018)。この成果は、実験と数理モデル、数値シミュレーションが有機的に融合することで初めて生まれるものであり、当初の予定よりも早く成果が結実した。また、バイオミメティクスの応用テーマであるマイクロロボットの開発にも成功し(Morita, et al., Physical Review E, 2018a, b)、当初の計画以上に研究が進展していると判断した。

Strategy for Future Research Activity

本研究では、微生物の力学環境に着目し、ナノスケールから細胞スケールに駆動力などの物性値を渡し、細胞スケールからメゾスケールに走性などの細胞応答を、メゾスケールからマクロスケールに応力や拡散などのテンソル量を渡すことで、ナノスケールからマクロスケールに至るまでの微生物懸濁液の振る舞いを記述する革新的な解析プラットフォームを構築する。
平成31年度は、主に細胞スケール、メゾスケール、マクロスケールの研究課題、および応用課題に取り組む。具体的には、以下の課題に取り組む。1.ベシクルの脂質二重膜上を移動する膜タンパクの移流拡散現象の解明。2.繊毛の協調運動による微生物の遊泳メカニズムの解明。3.微細藻類の重力感知メカニズムの解明。4.界面における微生物の振舞いと、生物学的な応答の解明。5.ゼブラフィッシュ腸内の輸送現象とフローラの解明。6.遊泳微生物を模倣したマイクロロボットモデルの開発
