Outline of Annual Research Achievements |
We attempted to trap the unknown E3 ligases for CRY1 protein, which is involved in regulating the circadian rhythms. We identified several candidate E3 ligases responsible for modifications of CRY1 by using our synthetic E2-SUMO probes in cell lysates for photo crosslinking. We are working on the validation of our hits with our collaborator, Dr. Tsuyoshi Hirota using siRNA knock downs of the candidate genes. We continued to synthesize new E2 conjugating enzymes. Also, we worked on the preparation of Ubiquitin and Ubiquitin-like proteins that can trap E2 conjugating enzymes for pulled down assays.
