含ホウ素ナノ粒子による薬物送達と中性子捕捉療法を組み合わせたがん治療法の開発

Outline of Annual Research Achievements

ホウ素中性子捕捉療法(BNCT)によるがん治療では、腫瘍への効率的かつ選択的なホウ素製剤送達システムを開発する必要がある。先行研究では、ホウ素製剤の生体環境下における分散性や腫瘍組織への選択性に問題があり、BNCT による治療効果はさほど高くなかった。
そして今回、まず、ホウ素を高濃度で含む含ホウ素ナノ粒子(B-NP)を親水性ポリマーであるポリグリセロール(PG)で被覆することにより B-NP-PG を合成し、その構造を確認するとともに、その高い水分散性を確認した。
次に、これを担がんマウスに導入し、中性子線を照射した後、マウスの腫瘍サイズを約1カ月間モニターしたところ、顕著な腫瘍成長抑制効果が見られた。特に、健康な状態を保ちながら、腫瘍が完全に消失したマウスも何匹か認められ、優れたがん治療効果が確認された。一方、コントロールでは腫瘍サイズが加速度的に大きくなっていった。
さらに、マウスの体重変化について見てみると、全てのマウスが約1ヶ月間、体重の緩やかな増加とともに健全な状態を維持した。一方、コントロールでは腫瘍サイズの増大に伴う体重の顕著な増加が見られた。
最後に、生存率においては、コントロールで何頭かのマウスが亡くなっているのに対し、B-NP-PG を注入し、さらに中性子線を照射した全てのマウスが約1ヶ月間、生存しており、中性子線照射下での B-NP-PG によるがん治療効果の優位性が、生存率の比較においても明らかとなった。
