Preparation of super spin glass magnetic nanoparticles and biomedical application for hyperthermia therapy

Outline of Annual Research Achievements

Co-Zn ferrite に加え、様々な遷移金属を用いて磁気微粒子を作製し、交流磁化率測定を詳細に測定し、1 nmごとに大きく変化する磁気緩和現象を分析した。特にMn(マンガン)にZn(亜鉛)をドープしたMn-Zn ferriteでは、超常磁性かつ磁化の値の大きい理想的な系が得られた。Mn0.8Zn0.2の割合がDC磁場において最も磁化が大きく、この組成における交流磁化率虚数部の値からは、18 nmの粒径が体温付近で最も熱散逸効果が高いことが予想できた。実際に交流磁場中で微粒子の温度上昇を測定したところ、予想通り、18nmの試料が最も上昇し、13度の昇温を確認した。ヒト乳がん細胞を用いたin vitro実験でも、3種類の異なるがん細胞で顕著なハイパーサーミア効果を確認した。
さらにMRIの造影剤としての機能を調べるため、このMn-Zn系のMR特性を測定したところ、興味深いことに、温度上昇が最も高かった試料と同じ組成のものが、もっともMR特性が効果的であった。現在使用されているリゾビストの成分である鉄酸化物、Fe2O3, Fe3O4の緩和能と比較しても約2倍の効率であった。この同じ試料を用いて
がん細胞選択性をねらった表面修飾に関しては、PETの手法をヒントに、磁気微粒子にグルコースを修飾する試みを行った。1st trialとしてこれまでの作製方法を一新し、SiO2で包含する代わりにポリエチレングリコール(PEG)で覆うことをまず試みた。FT-IRや質量分析スペクトルの測定結果を踏まえて、PEGおよびグルコースの修飾に成功したことを確認した。
さらに新しい診断方法として、磁気微粒子イメージング(MPI)への応用の可能性を期待し、この方法に重要なパラメータである、第三高調波成分の測定を手掛けた。
