Swimming behavior of zooplankton and its dietary value for fish larvae

Outline of Annual Research Achievements

昨年度の研究で、魚のアラ(FWD)を用いたワムシ培養がプロバイオティクス添加によって安定することを示したので、これを継続して、今年度はプロバイオティクスの投与がワムシの生理機能や遊泳にどのような作用を示すのか検討した。またFWDを餌料として量産したワムシを仔魚に給餌して、その効果を求めた。
微細藻類のNannochloropsis oculataで培養しているワムシに2種のプロバイオティクス製品(PB1、PB2)を添加し、ワムシの遊泳行動に与える影響を検討した。その結果、ワムシの平均遊泳速度は0.6 (対照)、0.75 (PB1)、0.95 mm/s(PB2)で、PB1投与のとき最大速度1.6 mm/sを示した。プロバイオティクスを投与しない場合、ワムシはシヌオシティ値(sinuosity; 正弦係数)が大きく、絶えず方向転換を繰り返したが、PB1とPB2投与では逆に直線運動が多くなる傾向がみられた。
次にFWDを餌料としたSS型ワムシの量産培養を実施し、100L水槽内でキスSillago japonica仔魚の飼育実験を行った。孵化から10日令までの成長と生残は、対照(クロレラ給餌ワムシ)との間に有意差がみられなかったが、仔魚の全長はFWD区の方が大きくなった。FWD給餌とクロレラ給餌で飼育した仔魚の全脂肪酸中のDHA含量はそれぞれ35.2%、18.2%で、FWDが低コストかつ優れた餌料源になることがここでも確認された。生残した仔魚は、形態と乾燥重量の測定に用い、予定していた遺伝子解析に供試する量を確保できなかった。今後の研究に繋げるため、研究室でモデル魚として飼育しているマングロープキリフィッシュの全ゲノム解析を実施した。
