抗体凝集体による免疫原性関連シグナル誘導の物理化学解析と数理モデル構築

Outline of Annual Research Achievements

最終年度の本年度は、医療用蛋白質の凝集体のサイズに応じた適切で定量的な評価方法、サイズごとの凝集体の発生機構、凝集体の免疫系への影響、を総合的に分析、それぞれの結果の関係性を解析し、総合評価を行った。凝集体評価として適切な手法は、サブミクロンが定量的レーザー回折法、ミクロンサイズはフローイメージング法、であることが明らかとなり、さらに、ミクロンサイズの画像からニューラルネットワークを用いた機械学習により蛋白質凝集体と容器からの剥離物を分類することに成功した。また、シリンジ等に塗布されているシリコンオイルが溶液中に放出されると油滴となり表面に蛋白質分子が整列するとIL-6やTNFなどの炎症性サイトカインのリリースへとつながる事が示された。また、蛋白質が結合した油滴は樹状細胞に取り込まれやすい性質を有していることも明らかとなった。これらの結果は眼科領域でも重要であったため、海外の研究者からの問い合わせが多く、最終的に総説としてまとめるに至った。
また、熱量測定において、撹拌翼で蛋白質溶液を撹拌するとミクロンサイズの凝集体が経時的に発生し、標的分子との結合活性が低下することを見出した。この手法は、物理ストレスに対しても安定な最適組成のための加速試験の設定にも有用であるといえる。さらに、凝集部位の特定を水素重水素交換質量分析法により実施し、アミノ酸番号257~281中にありCH2に位置する疎水性のアミノ酸が加熱時に露出することで凝集へとつながる事を特定した。以上の研究成果から、凝集を原因から取り除く手段、免疫系へのインパクトが高い凝集の解明、など蛋白質凝集体の総合評価に成功した。
