Outline of Final Research Achievements

Monitoring molecular orientation is the key approach to study the spatial organization of molecular alignments within supramolecular complexes in living cells. Fluorescence polarization microscopy is a promising solution, but it is challenging to label target proteins with fluorescent probes. We have developed a genetically encoded probe for fluorescence polarization microscopy. Observation of living starfish embryos expressing our probe that specifically binds to actin revealed a novel 3D structure that had been escaping from previous observations.

Academic Significance and Societal Importance of the Research Achievements

開発に成功した新規標識法は汎用性が高く、この技術を利用することによって今後、蛍光偏光を利用した生命科学領域における計測や観測の試行の増加が見込まれる。また、この新規手法により、私たちのグループの場合の応用例のような、高い基礎学術的意義を有する知見が蓄積すれば、細胞生物学、生物物理学、発生生物学をはじめ、基礎生命科学から蛍光計測を利用する応用生命科学まで、広範囲の応用成果が見込まれ、その社会的意義は大きいと予想される。
