ユビキチン修飾による腸管恒常性維持機構の解明

Outline of Annual Research Achievements

本邦において炎症性腸疾患(IBD)の罹患者数は増加を続け、その有効な予知・予防法は存在せず、内科的治療に抵抗する難治症例の克服は世界的な課題である。腸管は腸管上皮を介して外界と接し、微生物や食餌成分と共生している。IBDは、その共生関係の破綻により過剰な免疫反応が惹起された結果、発症すると考えられている。本研究では宿主のユビキチン化修飾反応に着目することにより、腸管恒常性の維持機構を解明し、IBDの病因・病態を明らかにすることを目的とする。本年度は、病原体関連分子パターン(PAMPs)に反応して発現するタンパク及びその制御タンパクに着目し解析を継続した。ユビキチンに対する免疫沈降法では、あるPAMPs反応タンパク及びその制御タンパクそれぞれがユビキチン化修飾を受けることを認めた。CRISPR-Cas9法を用いてそれぞれの遺伝子欠損細胞を作成し機能解析を行った。制御タンパクについて、ユビキチン化推定部位の変異タンパク発現ベクターを作成し、遺伝子欠損細胞に発現させ機能を解析した。また、PAMPs反応タンパクについて、質量分析を用いてそのユビキチン化修飾部位を推定し、今後はユビキチン化修飾部位の変異タンパク強制発現系を用いて機能解析を行う予定である。さらにはPAMPs反応タンパクに対する免疫沈降を行い質量分析で解析した結果、PAMPs反応タンパクの発現を制御する新規のタンパクも複数同定した。次年度はPAMPs反応タンパクの新規制御機構の詳細を解明する予定である。

Reason

PAMPsに反応するタンパクの発現制御機構について、ユビキチン化修飾に着目した結果、新たなメカニズムを見出したため。この新規発現制御機構は、腸管恒常性維持の一翼を担う新たなメカニズムであると考えられる。現在まで、遺伝子欠損細胞や変異タンパク強制発現系による機能解析、タンパク質複合体の相互作用解析などを行い、概ね順調に進展していると考える。

Strategy for Future Research Activity

前年度に同定された新規の制御タンパクの遺伝子ノックダウンを行い、それら制御タンパクの機能を詳細に解析する。また、PAMPs反応タンパクにおける、ユビキチン化修飾部位の変異タンパク強制発現系により、そのユビキチン化修飾の機能を明らかにする。最終的に実験結果を取りまとめ、新たな腸管恒常性維持機構を明らかとし、IBDの新規治療戦略を提案する。
