Elucidation of mechanism underlying intrinsic atopic dermatitis induced by deficiency of sprabasin co-expressed in skin and intestine

Outline of Annual Research Achievements

血清IgE値が正常の内因性ADの原因は未だ不明であるが、我々はスプラバシンsuprabasin (SBSN) 欠乏が金属アレルギーに関連することを推定している。そこでSBSNノックアウトマウス(SBSN-/-マウス)を作成しこれまで解析を行ってきた。当該年度では、SBSN-/-マウスにおける接触過敏反応を中心に検討した。通常のハプテンである、塩化ピクリル、DNFB、オキサゾロンに対する接触過敏反応は、野生型マウスに比べ予想外に低下していた。またこれらのハプテンで感作したマウスの所属リンパ節T細胞の、in vitroでのハプテンに対するサイトカン産生能も低下していた。従ってSBSN-/-マウスでは何らかの免疫学的な理由があり、接触過敏症が低下することが考えられたが、その仔細は未だ解明できていない。次に、SBSN-/-マウスでの金属アレルギーの程度を検討するために、ニッケルを負荷したマウスでのニッケルに対する接触過敏症の程度を解析した。ニッケル14日間の経口飲料負荷でSBSN-/-マウスはニッケル濃度が野生型マウスに比べ有意に上昇することを見出し、このニッケル濃度が上昇したSBSN-/-マウスにニッケルの接触皮膚炎を誘導・惹起した。その結果、ニッケル負荷なしのマウスに比べて、有意に皮膚炎反応が亢進していた(Nakazawa S et al. Sci Rep 2020)。本研究により、SBSN欠乏がニッケルの血中濃度を高め、金属の接触皮膚炎の亢進を誘導することが明らかになった。SBSN欠乏は内因性ADの一つの原因になっていることが示唆された。
