Optimal ultraviolet wavelength for immune regulation and development of ultraviolet-LED phototherapy

Outline of Annual Research Achievements

平成29年度~30年度の成果において、iTreg(誘導性制御性T細胞)を誘導する波長解析では、290~320nm中で、310nmに遅延型過敏反応の抑制とTregの誘導が見られた。nTreg(内在性制御性T細胞)の誘導波長解析 抗原の投与していないマウス皮膚にリンパ節・脾臓でCD4+CD25+Foxp3+Neuropilin-1+細胞(nTreg)が、290~300nmで誘導が見られた。皮膚とリンパ節でTregが増加、Neuropilin1陽性のnTregであり、Treg関連の遺伝子におけるメチル化を調べたところ、脱メチル化がみられ、安定した抑制機能のあるTregが誘導されることを明らかとなった。臨床レベルでのTreg誘導解析では、乾癬患者のTregの抑制能の低下は、PUVAバス療法後においてほぼ正常へ回復したことから、PUVAバスによるTregの機能回復が示した。照射開始 (PUVAバス)から5回ごとに、3つの分画を調べたところ、aTreg (CD4+CD25+Foxp3highCD45RA-)が、明らかに上昇することが明らかとなった。比較的早期にaTregが上昇し、rTregも同様に誘導され、非抑制性 Tregの減少もみられた。マウス皮膚で、Tregが誘導されたことから、ヒトでも同様のTregの誘導がわかるかどうかを確かめるため、皮疹血として採取したわずかな全血から、細胞解析ができるかどうかの検討を行った。マルチカラーでのFACS解析、さらに高速セルソーターで、CD4/ CD8を分離することも成功した。末梢血に比べ、皮疹血では、CD4およびCD8のpopulationに差があり、制御性T細胞の分離にも成功し、RNAシーエンスの解析を行っている。
