Outline of Annual Research Achievements

我々は、「象牙芽細胞下層と歯髄中央部の血管周囲に前駆細胞/静的幹細胞(長期ラベル細胞:LRCs)が配置しており、歯髄の恒常性を維持している」という仮説を提唱した。本研究課題では、ドキシサイクリン(Dox)投与ですべての細胞をGFPでラベルした後、細胞分裂回数に応じて細胞ラベルが減弱するノックインマウス(TetOP-H2B-GFPマウス)を用いた。H2B-GFPラベル維持細胞(H2B-LRCs)は歯髄中央部に加え象牙芽細胞下層に分布していた。IGFBP5/Igfbp5発現は生後4週で歯髄中央部と象牙芽細胞下層に見られ、これらは非対称分裂する歯髄幹細胞/前駆細胞と共局在し、歯根膜にも局在した。外的侵襲後に歯髄幹細胞/前駆細胞がIGFBP5を発現することで、アポトーシスから逃れ、歯髄内に維持されることから、歯の発生や外的侵襲後の歯髄治癒過程において、IGFBP5が歯髄幹細胞/前駆細胞の生存に重要な役割を果たすことが示唆された。プロテオミクス解析では、3週齢ICRマウスの第1臼歯の象牙芽細胞層、象牙芽細胞下層、歯髄中央部から、切歯形成端の上皮幹細胞ニッチであるapical budと下層の歯乳頭から組織を採取した。象牙芽細胞特異的タンパク質156、象牙芽細胞下層特異的タンパク質183、歯髄中央部特異的タンパク質76を同定した。三者間で988、二者間で76~149のタンパク質を共有していた。apical bud部の上皮と間葉は、それぞれ258、318の特異タンパク質、1350の共通タンパク質を同定したが、現在タンパク質の局在を解析中である。さらに、apical budにおいて、H2B-LRCsが低酸素状態にあり、微小環境の酸素が幹細胞の増殖制御に重要な役割を果たすことを明らかにした。また、接合上皮幹細胞がapical budの幹細胞と同じ動態を示す可能性を示唆した。
