Outline of Annual Research Achievements

セントロメアは、染色体分配に必須なゲノム領域である。染色体の正確な分配は、生命の維持にとって必要不可欠であるため、セントロメアを中心とした染色体分配機構の解明を目指した研究はきわめて重要である。我々は、20年以上にわたり、セントロメアの分子基盤の解明を目指した研究を精力的に行ってきており、国内外から高い評価を受けてきた。本研究では、我々のこれまでの研究をベースに、それをさらに発展させて、セントロメアの形成機構を解明することを目的として研究を進めた。具体的な計画では、I) 細胞周期に依存したセントロメア構成タンパク質ネットワークの理解、II) セントロメアが形成されるゲノム基盤の理解、III) 電子顕微鏡を活用したセントロメアタンパク質複合体の構造基盤の理解の3計画の研究を平行して進めた。特に、2020年度に進める予定であったMis12複合体とCENP-Tとの関連研究が、コロナウイルスの影響で共同研究が遅れたものの、2021年9月までに研究を行い、CENP-TにおけるMis12複合体の意義を明らかにでき、論文発表に至った。また、関連して、セントロメアが形成されるゲノム基盤の理解を目指した研究では、新規CENP-A取り込み機構を明らかにし、電子顕微鏡を活用したセントロメアタンパク質複合体の構造基盤の理解の研究においては、KNL2とCENP-Aヌクレオソームの複合体構造を明らかにするなどの成果が得られた。
