Outline of Annual Research Achievements |
DNA adductome analyses of Hep G2 cells exposed to the monoaromatic chemicals, safrole and monoaromatic amines were conducted including DNA adductome mapping and LC/ESI-MS/MS CID analyses of DNA adduct candidates. Research results related to safrole exposure were finalized and submitted for peer review. Methods were established (1) for Hep G2 cell viability assays, (2) for in vitro monoaromatic chemical reaction assays with S9, extraction, and LC/ESI-MS/MS analyses, (3) for adduct synthesis of a monoaromatic amine DNA adduct, and (4) for cultivation of a human bladder cell line. Methods for "RNA adductomics" were developed and some experiments were completed.

Strategy for Future Research Activity |
(1) Conduct DNA adductome analyses using bladder cell line, (2) continue syntheses of DNA adduct standards and apply to DNA adductomics, (3) develop high resolution mass spectrometry HRMS approaches to apply to adductomics, (4) continue development of "RNA adductomics".
