Outline of Final Research Achievements |
The pathogenic mechanism of renal angiopathy in prostacyclin (PGI2) deficient mice is unclear. In this study, we aimed to clarify the onset time and signal transduction pathway of renal disorder using PGI2 synthase gene-modified mice. In addition, the relationship with adipose tissue was examined using cultured adipocytes. It was revealed that PGI2 deficiency increased the expression of inflammatory cytokines in renal tissue by 28 days after birth. It was also suggested that the signal transduction system via PGI2-PPAR-delta may be important for the maintenance of renal function. The increased expression of inflammatory adipocytokines in adipocytes due to inflammation was suppressed by n-3 polyunsaturated fatty acids, and it was expected that the onset of renal vascular disorders could be prevented.

PGI2の欠損あるいは産生レベルの低下が、腎臓に炎症を起こしやすい状況を生じることや腎機能の維持にPGI2-PPAR-deltaを介するシグナル伝達機構が重要な役割を果たす可能性を示唆する本研究結果は、近年我が国において問題となっている生活習慣病による慢性腎臓病や血管の老化に伴う腎機能低下の病態改善、発症・進展予防の研究の基盤として役立つものである。
