Outline of Final Research Achievements

In this study, based on the BioID enzyme labeling technique using a biotin ligase mutant (BirA), we searched for essential molecules involved in muscle atrophy via heat shock proteins (HSPBs). As a result, we succeeded in identifying several molecules that are included in the cytoprotective function of HSPBs to suppress muscle atrophy caused by endoplasmic reticulum stress. This result suggested cellular stress protection via the Polo-like kinase 2 (PLK2)/HSPB5 pathway in the endoplasmic reticulum stress response of muscle cells.

Academic Significance and Societal Importance of the Research Achievements

高齢者では、筋力及び筋肉量が低下し、持続的な運動負荷により改善が期待される。しかし、高齢者の効果的な筋萎縮予防の実践には、過度な運動負荷による身体への負担が懸念される。本研究では、HSPBsに着目し、筋萎縮の抑制に効果的なHSPBsの機能を明らかにした。今後、HSPBsの発現増加に着目した栄養成分を探索することで、過度な運動負荷に頼らない予防法の考案に繋がることが期待される。
