Outline of Final Research Achievements |
In this study, one of the effector complesx of Rab GTPase sec4, Exocyst complex, was focused to analyze for the initiation of prospore membrane formation during sporulation in Saccharomyces cerevisiae. We have constructed auxin-inducible degron mutants for each subunit of Exocyst complex and analyzed for prospore membrane formation. Although each of them did not show complete sporulation defective phenotypes, each mutants showed reduced sporulation efficiency, suggesting the Exocyst complex functions in prospore membrane formation.

Academic Significance and Societal Importance of the Research Achievements |
本研究で明らかにする膜構造形成の分子機構は、出芽酵母の前胞子膜のみならず様々なオルガネラにおいて、さらには種を超えて存在する。よって本研究により得られる知見を足がかりとして細胞内膜交通の基本的な分子基盤が明らかになることが期待される。本研究ではExocyst複合体の前胞子膜形成への寄与を明らかにした。他生物におけるポストゴルジ小胞同士の融合による新規膜構造形成としては精子頭部の先体や植物細胞の細胞分裂時の細胞盤形成が挙げられ、Exocyst複合体はこれら膜構造形成にも同様に機能することが考えられる。
