Outline of Annual Research Achievements |
Preliminary analysis of DNA base modification patterns during polyp-to-jellyfish transition has been performed using PacBio RS II and NanoPore sequencers. Data analysis showed that the increased sequencing depth (~150-fold genome coverage)is needed in order obtain statistically reliable results. In case of PacBio RS II machine this level of coverage is prohibitively expensive. Taking into consideration that PacBio Sequel II sequencer will be operational in our institute in FY2020 we decided to delay the experiments until the new equipment will become available.

Reason
In FY2020 new PacBio Sequel II sequencer will start to operate in OIST with ~100 fold mode data that PacBio RS II machine for the same price (initially planned sequencer was PacBio RS II). Thus we decided to extended the grant period for FY2020 in order to obtain the desired results in more efficient way.

Strategy for Future Research Activity |
I needed to change the research plan (extended grant period for 1 year) because much more efficient research equipment (PacBio Sequel II sequencer) will be available starting from July, 2020. Otherwise the project will proceed according to the initial plan.

Causes of Carryover |
According to the initial research plan we intended to used PacBio RS II sequencer to study epigenetic changes during polyp-to-jellyfish transition. In FY2020 the next generation PacBio sequencer (Sequel II) will start to operate in OIST. This new machine produces ~100 fold mode data for the same price. In order to use the research budget in the most efficient way I decided to shift the project for FY2020. Remaining budget will be used for PacBio sequencing as originally planned, but using a more efficient Sequel II machine.
