Reversible and irreversible damages of hepatocyte after DAA treatment for HCV carriers

Outline of Annual Research Achievements

HCVの長期間にわたる持続感染による肝炎による細胞障害、組織障害の分子機構と治療によるその可逆性についてはよくわかっていない。本研究ではHCV感染モデルを用いて、HCV感染肝障害の微細構造変化および遺伝子発現変化を解析し、DAA治療によるウイルス排除後の肝障害改善に資する基盤的研究を実施する。ウイルス培養系において、未感染細胞、感染細胞、排除後細胞を区別できる実験系を構築するためにスイッチング発現系を利用する。平成31年度は単クローンHuh751-RG細胞から、RFP/GFP組換えが起こるウイルス感染を経験したHCV感染細胞、HCV排除細胞へと一連の過程の細胞を得た。令和元年度は、感染後のHCV産生が安定している3個の単クローン細胞に注目し、それらのHCV未感染細胞、HCV感染細胞、HCV感染後排除細胞について、細胞内の微細構造を観察した。感染細胞では二重膜構造や脂肪滴構造の出現、ミトコンドリア構造の異常が観察された。排除細胞においてはこれらの構造変化は感染前の状態に回復していた。この構造変化はHCV感染による宿主の遺伝子発現変化によるものと考え、HCV感染後、HCV感染から排除後の遺伝子発現の変化をRNA-seqで網羅的に解析した。HCV感染により約580遺伝子、その後のHCV排除により約360遺伝子に発現変化があった。これら遺伝子の中にはHCV排除により感染前の状態に回復する遺伝子、回復しない遺伝子があった。これらの遺伝子のうち感染により遺伝子発現が増加する20遺伝子(排除後に回復しないものが12遺伝子)、減少する12遺伝子(排除後に回復しないものが4遺伝子)に注目して解析を進める。中でもウイルス排除後に回復しない遺伝子に関しては、不可逆的な肝組織変化に関与すると考えられ、解析を進めることで肝組織異常の改善につながる情報になり得る。
