Outline of Annual Research Achievements |
ヒト食道癌組織におけるTRPV2の発現レベルを解析したところ、TRPV2高発現群の予後が有意に不良であった。TRPV2を高発現する、ヒト食道癌細胞株:TE15、KYSE170を用いて、TRPV2 siRNAのトランスフェクションを行い、細胞増殖抑制効果・アポトーシス増強効果、細胞遊走・浸潤能抑制効果を確認した。また、TRPV2のノックダウンにより、EMT関連蛋白であるSnailの低下と、Apoptosis関連蛋白であるCleaved Caspase3の増加が確認された。更に、TRPV2 siRNAを導入したTE15細胞株の遺伝子発現変化をmicroarrayにより網羅的に解析した。Ingenuity Pathway Analysisの結果では、Wnt/β-カテニンシグナル伝達系に遺伝子発現変化を認めた。これらの研究成果は英文論文にまとめて発表した(Sci Rep. 2019)。 更に、食道扁平上皮癌細胞株TE4、TE8から癌幹細胞を抽出・培養し、その遺伝子発現変化をmicroarrayにより網羅的に解析した。TE-4、TE-8由来癌幹細胞株に共通して、TRPV2とともに、ATP2A1, ATP2A3, ORAI3などの、他のカルシウムイオンチャネルやトランスポーターが高発現することを見出し、その阻害剤thapsigarginの癌幹細胞増殖抑制効果を確認した。 一方で、消化器癌におけるANO9 (Ann Surg Oncol. 2020)、LRRC8A (Am J Pathol. 2019)、Sodium iodide symporter (Gastric Cancer. 2019)などのイオン輸送体の発現機能解析・臨床病理学的意義を検証するとともに、低浸透圧刺激後の温度・イオン輸送体・水輸送体を介する細胞容積調節機構を解明した(Int J Oncol. 2019)。
